Different methods, i.e. basic method, high salt precipitation, CTAB precipitation, low pH extraction medium with high salt, were used to extract genomic DNA from the needle leaves of Pinus kesiya var langbianensis. The DNA samples obtained by the above methods were tested by agarose gel electrophoresis, restriction digests and PCR. The difference in yield, quality of the DNA samples was compared, and high salt precipitation was considered to be a fast, inexpensive and reliable technique among the four methods.
